Pros: GPS interface, voice prompts, "radar" searches, accuracy of routing, database
Cons: User interface, creation & editing of routes
I actually tried all three of these programs, looking for the best solution for a cross-country trip. MapPoint and Streets and Trips are virtually identical in database and interface with MapPoint having additional features I liked for their business applications (geographic mapping of databases, etc.). I found the Street Atlas interface incredibly hard to use. Scrolling the screen (a simple click and drag in the Microsoft products) takes multiple mouse clicks and waiting for the screen to refresh. Editing routes also seemed much harder with Street Atlas.
It may come down to purely personal preference, but I found Street Atlas so unusable while driving that I gave away my copy. I now use Streets and Trips on the road and MapPoint at work.
